The long-term goals of this project are to elucidate the cellular and molecular underpinnings of light-sensing in native cells that utilize the photopigment melanopsin. The chicken retina presents itself as an interesting and valuable system to study melanopsin transduction due to the abundant expression of this protein in the inner nuclear layer possibly in bipolar and horizontal cells. The functional role of melanopsin in these retinal layer is completely unknown which opens the possibility of uncovering a completely novel light sensor. More important this organism may provide clues on the evolution of different lineages of photoreceptors and their respective signaling cascades.en
oaire.objetivesSpecific Objectives : 1. Develop a preparation of functionally viable cells from the retina of chicken embryos, isolated by means of enzymatic treatment and mechanical dispersion.en
oaire.objetivesSpecific Objectives : 2. Determine which of the defined cell types present in the inner nuclear layer of chicken embryo retinas express melanopsin.en
oaire.objetivesSpecific Objectives : 3. Use a fluorescence assay to establish the presence of retinal in melanosin-expressing cells, which would suggest that melanosin forms a functional photopigment.en
oaire.objetivesSpecific Objectives : 4. Utilize patch-clamp recordings to examine whether melanopsin-expressing cells are intrinsically photosensitive.en
oaire.objetivesSpecific Objectives : 5. Detect and localize key signaling molecules of the phosphoinositide pathway in the chicken retina, using Western blot analysis, and isolated-cell immunocytochemistry.en
oaire.objetivesSpecific Objectives : 6. Ascertain the functional role of key downstream elements of the cascade, by means of single-cell electrophysiology in conjunction with Ca fluorescence measurements and pharmacology.en
