dc.description.abstractScientists at CIAT and Universidad del Valle have identified wild cassava accessions resistant to green mite. In order to foster the introgression of the resistance trait into farmer-preferred cultivars, the research project proposes to undertake a molecular characterization of the mechanisms underlying the green mite resistance in the wild cassava accessions. The recent release of the cassava genome allows the implementation of advanced label-free protein quantification to characterize the proteome and its modulation upon arthropod attacks. The system, green mite - cassava, will be investigated with the resistant and susceptible accessions for the system. In addition, it is intended to evaluate whether the resistance of the selected cassava accessions is induced or constitutive. To do so, a comparison of pest performance on (1) plants previously unexposed to arthropod attack with (2) plants previously challenged by arthropods, will be performed. For the complete experimental set, sampling at several key time points during arthropod establishment and development will allow deciphering the constitutive and inducible proteome differences. The label-free protein quantification will generate a list of gene candidates whose expressions relate to arthropod resistance for the system under investigation. Specific qPCR primers will be developed and transcript quantification of the selected gene candidates will be performed on the leaf material sampled during the aforementioned entomological experiments. For each candidate gene, the following transcript measurement will be generated: relative expression between susceptible and resistant accessions, time course expression analysis, and expression level.spa
